Molecular interaction affinity

LSPR Molecular Interaction System

Affinity is an important parameter to determine the intermolecular interaction, and an important indicator to understand the discovery and screening of molecules and drugs. In addition to affinity, there are two important indicators (Ka, Kd) for the evaluation of intermolecular interaction. For the evaluation of efficacy and the stability of biomacromolecules and their complexes, it is necessary to conduct a comprehensive study from the aspects of dynamics, thermodynamics and thermal stability, including the intermolecular binding, the speed of binding, the strength of binding, and the binding mechanism.

Based on LSPR platform, Yanjin Bio provides corresponding molecular interaction analysis and testing services for biomedical R&D and scientific research clients, which can achieve qualitative and quantitative analysis of interactions of proteins, antibodies, antibody fragments, polypeptides, DNA/RNA, nanomaterials and small molecular compounds, and can also be used to fish and screen unknown targets based on LSPR Selex technology for cell lysate, nucleic acid library, etc.

Detection scope: detection scope includes protein protein, antibody antigen, protein small molecule, antibody polypeptide, protein DNA, DNA DNA, protein nanomaterials and other molecular interactions; In addition to proteins and antibodies, cell lysates and bacteria can also be combined with LSPR-MS fishing mass spectrometry to identify regulatory proteins; It can also be widely used in drug screening, medical diagnosis, environmental food detection, blood marker detection and other fields.

Product requirements:

1 Ligand protein (marker chip)

1.1. Purity greater than 90%

1.2 If the concentration is greater than 0.1ug/ul (as high as possible), about 10ug shall be provided.

1.3. It is required to provide the isoelectric point of ligand protein

1.4. Try to avoid Tris and other reagents with amino groups in ligand protein buffer solution

1.5 Avoid particle or flocculent sedimentation

2. Analyte (mobile phase)

2.1. Purity greater than 80%

2.2 If the concentration is greater than 0.1ug/ul (as high as possible), it is better to provide freeze-dried powder. According to the sample condition, provide about 10-100ug.

2.3. Provide accurate molecular weight, concentration and solvent composition

2.4 Solvents should preferably not contain organic compounds, such as DMSO glycerin. If it is necessary to add organic solvent, try to provide the analyte with high concentration, and the content ratio can be reduced by dilution.

2.5. Try not to contain trehalose, BSA, sucrose and other protective agents. If it is necessary to add, it is necessary to provide an accurate concentration, as well as a high concentration of analyte containing stock solution as far as possible, so as to reduce its content ratio by dilution. After dilution, the concentration of antigen antibody protector in the final concentration of the sample can be ≤ 0.01%.

Product advantages:

1. Technical advantages: Yanjin Biology focuses on the development, application and technical services of molecular interaction methodology, providing customers with integrated solutions from SPR technical services to diagnostic product development and molecular interaction instrument.

2. High throughput and high sensitivity detection: Yanjin Bio can detect multiple samples at the same time, with the detection concentration range of 0.1nM-1mM samples, and the affinity detection range of fM mM

3. Wide application range: it can withstand various solution environments and directly detect crude samples, such as serum, tissue lysate, viral macromolecules, etc.

4. Meet different experimental requirements: 10 kinds of chips are used to detect the interaction between different ligands and analytes, including blank gold nanoparticles, COOH, NH2, NTA, Streptavidin, Biotin, etc.

5. Strong anti-interference ability: the detection is not affected by temperature, buffer refractive index and other external environments.

6. Real time analysis without marking: save time and materials, and monitor in real time.

7. Powerful function: it can be used to analyze the interaction between large and small molecules such as proteins, nucleic acids, lipids, polypeptides, nanomaterials, etc.

8. It is widely used in biochemistry, drug development, tumor research, immunology research, antibody engineering, etc.
